چکیده

Background Due to a lack of antioxidant enzymes in plasma, plasma proteins are preferential targets of oxidant injury. Reaction of chlorinated oxidants (e.g. hypochlorous acid), generated from activated neutrophils, with plasma albumin gives rise to formation of an oxidized protein fraction, termed advanced oxidation protein products (AOPPs). AOPPalbumin is a potent high-density lipoprotein (HDL) receptor antagonist, blocking HDL association and reverse cholesterol transport (RCT). However, it is not known whether structural alterations and/or an increase in negative charge (through oxidation of positively charged lysine residues on albumin) are required for high affinity binding to SR-BI.

Fluorescein isothiocyanate specifically modifies lysine 338 of cytochrome P-450scc and inhibits adrenodoxin binding.

Treatment of cytochrome P-450scc with fluorescein isothiocyanate (FITC) resulted in covalent labeling with 1.0 +/- 0.1 eq of FITC. Reverse-phase high performance liquid chromatography of tryptic and chymotryptic digests of the labeled protein revealed that a single FITC-labeled peptide accounted for 75% of the label. This peptide was found to be specifically labeled at lysine 338 by amino acid ...

Inhibition of hepatic scavenger receptor-class B type I by RNA interference decreases atherosclerosis in rabbits

OBJECTIVE Scavenger receptor-class B type I (SR-BI), the receptor for HDL-cholesterol, plays a key role in HDL metabolism, whole body cholesterol homeostasis, and reverse cholesterol transport. We investigated the in vivo impact of hepatic SR-BI inhibition on lipoprotein metabolism and the development of atherosclerosis employing RNA interference. METHODS Small hairpin RNA plasmid specific fo...

An intracytoplasmic injection of deionized bovine serum albumin immediately after somatic cell nuclear transfer enhances full-term development of cloned mouse embryos.

In mouse somatic cell nuclear transfer (SCNT), polyvinylpyrrolidone (PVP) is typically included in the nuclear donor injection medium. However, the cytotoxicity of PVP, which is injected into the cytoplasm of oocytes, has recently become a cause of concern. In the present study, we determined whether bovine serum albumin deionized with an ion-exchange resin treatment (d-BSA) was applicable to t...

Cross-sectional examination of metabolites and metabolic phenotypes in uremia

BACKGROUND Although metabolomic approaches have begun to document numerous changes that arise in end stage renal disease (ESRD), how these alterations relate to established metabolic phenotypes in uremia is unknown. METHODS In 200 incident hemodialysis patients we used partial least squares discriminant analysis to identify which among 166 metabolites could best discriminate individuals with ...

Plasma lipid transfer proteins.

The plasma lipid transfer proteins mediate the transfer and exchange of phospholipids and neutral lipids between the plasma lipoproteins. The cholesteryl ester transfer protein (CETP) and the phospholipid transfer protein (PLTP) are members of the lipid transfer/lipopolysaccharide binding gene family. The CETP contains binding sites for cholesteryl ester and triglycerides and probably acts by a...
